Hi I am considering buying a 2005 XK8 with 45k on the clock, at this mileage could there be any issues with the differential. The reason I ask this is because I'm on my third diff on my S Type and i'ts only done 77k. I know by reading this and other forums that the company that was manufacturing Jag differentials at a certain time didn't do a very good job! And is there other thing that I should be looking out for.

Check for N/S drive shaft oil seal failure/leaks. A leak usually means the diff has an issue.
Comparable car, XJR 350. I`d immediately have diff oil changed if you buy the car. Diff oil is cheap, diffs are not! I`ll be changing diff oil again at the next 60k service interval. I imagine the service schedule for your proposed car is every 90k? Far to long in my experience. You should see what comes out at 60k. Friction protection is zero! Nuff said?

Changing all the fluids in a newly purchased car is never a bad investment in its future, though access to the diff in an XK is not easy; there's no drain point so a suction pump has to be used through the filler plug.
I've followed/participated in two other Jaguar XK8/R Forums over the last ten years and would say, based on that, that XK8/R diff problems don't make the top ten of concerns.
Always first is rust. Even with an enviable low mileage of 45k, the underside needs a thorough examination. I think I caught all mine before it got serious, but areas to examine include:
The floorpans in way of the doubler plates fitted to assist manufacture by Jaguar, but notorious water/rust traps.
The front steel crossmember, its rear face especially.
The mounting points for the A Frame of the rear suspension
A difficult to reach/see, but highly susceptible area, is where the front inner wings pass behind the uprights of the alloy subframe
With a torch , check out the chassis/inner wing areas between the headlights and the suspension turrets.
Obvious really, but make sure all the electrical items work correctly, particularly
If you can get a comprehensive diagnostic scan of the car's systems to prove no hidden faults it would help put your mind at rest.
